No, it is not necessary to give your campaigns certain settings to enable Rotations, but it can only help. :) 

Best Practice:

For those networks (below) that allow you to set a delivery method for your ads, we recommend that you set them to "rotate evenly". This will ensure each ad has been given the same opportunity to be served, and allow Rotations to run more effectively.

Networks with "rotate evenly" options: 

AdWords

Step 1: Sign into AdStage and select an Adwords Campaign
Step 2: Find the Settings tab and click Edit Settings
Step 3: Find Delivery Method, under Bidding & Budget and expand the section
Step 4: Select "Standard: Show ads evenly over time"

Bing 

Step 1: Sign into AdStage and select a Bing Ad Group
Step 2: Find the Settings tab and click Edit Settings
Step 3: Under Advanced Settings, Find Ad Rotation
Step 4: Select "Rotate ads more evenly"

LinkedIn 

Step 1 - Sign into your LinkedIn account, and select a campaign
Step 2 - Find and click the "gear icon" of the campaign
Step 3 - Click Rotate ad variations
Step 4 - Select "Rotate variations evenly"

Networks without "rotate evenly" options:
